Ameliorative Effects of Apigenin and Vitamin E on Biochemical and Histopathological Changes in Rat Model of Cyclophosphamide-Induced Nephrotoxicity

Abstract

This study aims to evaluate the potential nephroprotective effects of apigenin, vitamin E and their combination against cyclophosphamide-induced renal injury in male rats. Forty male albino rats were randomly divided into five groups. Group 1 received vehicle (corn oil and glycerin) and served as the normal control, Group 2 received cyclophosphamide (CYP)150mg/kg, Group 3 received vitamin E 100mg/kg plus CYP, Group 4 received apigenin 20mg/kg plus CYP, and Group 5 received a combination of apigenin and vitamin E plus CYP. Group 2,3,4 and 5 were treated intraperitoneally on day 17 with a single dose of cyclophosphamide. Serum levels of malondialdehyde (MDA), total antioxidant status (TAS) and β2-microglobulin (B2MG) were assessed. Caspase-3 expression was also performed to determine apoptotic activity. The results show that the Cyclophosphamide administration significantly increased MDA and β2-microglobulin levels and enhanced caspase-3 expression, while significantly reducing total antioxidant status compared to the control group. Treatment with vitamin E and apigenin individually attenuated oxidative stress and apoptotic changes. Notably, the combination therapy demonstrated the most pronounced improvement in antioxidant status, reduction in lipid peroxidation and suppression of caspase-3 expression. Apigenin and vitamin E exert protective effects against cyclophosphamide-induced renal injury through modulation of oxidative stress and apoptosis. Their combined administration showed superior efficacy, suggesting a synergistic antioxidant and anti-apoptotic mechanism.
