Exploring the mechanism of action of Huoxue Quyu San hot package in intervening in sequelae of pelvic inflammatory disease based on network pharmacology and molecular docking

Abstract

Abstrsct: Objective: To investigate the therapeutic mechanism of Huoxue Quyu San Reyan Bao in treating sequelae of pelvic inflammatory disease (SPID) using network pharmacology and molecular docking. Methods: Active components and targets of Huoxue Quyu San were obtained from TCMSP, HERB, and UniProt. SPID-related targets were retrieved from GeneCards and OMIM. Common targets were identified via Venny 2.1.0, and a herb-component-target-pathway-disease network was constructed using Cytoscape 3.10.0. A PPI network was built using STRING to screen core targets. GO and KEGG enrichment analyses were performed using DAVID. Molecular docking was conducted using AutoDock Vina 1.5.7 to validate binding affinities. Results: A total of 170 active components and 154 common targets were identified. Key components included quercetin, apigenin, baicalein, luteolin, and daidzein. Core targets were AKT1, TNF, IL-6, INS, IL-1β, and TP53. Involved pathways included cancer, TNF, PI3K-Akt, AGE-RAGE in diabetic complications, IL-17, lipid/atherosclerosis, C-type lectin receptor, and fluid shear stress/atherosclerosis pathways. Molecular docking showed strong binding affinities. Conclusions: Huoxue Quyu San treats SPID by modulating TNF, PI3K-Akt, and AGE-RAGE pathways, exerting anti-inflammatory, antioxidant, microcirculation-improving, and apoptosis-inducing effects.
