Abstract

Radiotherapy and chemotherapy are widely employed in cancer treatment but frequently induce intestinal toxicity, which significantly compromises patients’ quality of life and limits therapeutic efficacy. Polyphenols, a diverse class of natural compounds with broad biological activities, have demonstrated protective effects against radio/chemotherapy‐induced intestinal injury through antioxidation, anti‐inflammation, DNA damage repair, and microbiota modulation, etc. However, the limited stability, bioavailability, and targeting efficiency of polyphenols hinder their oral use in clinical practice. This review summarizes the mechanisms and oral delivery challenges of polyphenols. Furthermore, it highlights recent advances in oral delivery systems designed to address the oral limitations of polyphenols. Challenges in developing such systems and potential future directions are also discussed, aiming to provide new perspectives for oral polyphenol formulations to prevent/treat radio/chemotherapy‐induced intestinal toxicity.
